Introduction:

Our ready-to-use normal tissue Western blot strips are an innovative solution to the pre-made Western blot sample content conundrum - not getting all or exactly the right samples for your analysis at an affordable price. Buy as few or as many, in any combination, to suit your experimental needs. Our blots are prepared using 4-20% pre-cast 1D-PAGE gradient gels for maximum protein separation and resolution. Gel to gel loading and running are constantly monitored for consistency and reproducibility to maintain high quality standards. These products are made using supported PVDF membranes for high protein binding capacity and are extra strong to resist tearing and permit easy handling. Simply align the strip to the marker key provided on the product data sheet to determine the molecular weight of your target. Each strip comes individually packaged in a convenient 2 ml incubation chamber, ready-to-use and minimizing the amount of reagents and antibodies required.T

 

These products are ideal for protein biomarker analysis platform and are ideal for biomarker identification / screening, antibody detection / characterization and protein expression analysis. Target detection may be accomplished using a number of detection systems including color development, enhanced chemiluminescence (ECL) and fluorescence.

Instructions:

1) Pre-wet PVDF strip in 100% methanol.

2) Rinse pre-wetted membrane several time in TBS-Tween 20 or PBS-Tween 20 (i.e. Wash Buffer).

3) Transfer to protein blocking buffer.

4) Incubate with gentle rocking/shaking for 60' at room temperature.

5) Add primary antibody to the concentration/dilution factor recommended by the manufacturer. Incubate with gentle rocking/shaking for 60' at room temperature; alternatively incubate overnight at 4o C.

6) Wash membrane in Wash Buffer 6 times, 5' each at room temperature.

7) Repeat steps 3-7 using recommeded secondary antibody.

8) Detect signal using according to manufacturer's detection reagents/kit.

9) Determine the molecular weight of the target protein by aligning the top of the PVDF membrane with the top line of the diagram on the last page of the product datasheet.
